In 2017, he made his film debut as Vincent Pepe in the Malayalam-language crime drama film Angamaly Diaries. His co-stars for the project included Anna Rajan, Tito Wilson, and Sarath.

For his performance in Angamaly Diaries, Antony Varghese bagged the Filmfare Award South in the “Best Malayalam Male Debut” category in 2018.

He brought to life the character of Jacob Varghese in the 2018 Malayalam-language thriller film Swathanthryam Ardharathriyil (also spelled Swathandriam Ardharathriyil).

Antony Varghese co-starred as Antony (Kalan Varkey’s assistant) in the critically-acclaimed 2019 Malayalam-language independent action thriller film Jallikattu. The film was selected as the Indian entry for the “Best International Feature Film” at the 93rd Academy Awards and is widely regarded as one of the defining movies of the New Wave Movement.

In 2021, Antony was cast as Lali in the Malayalam-language action thriller film Ajagajantharam.
